2000 2.7L - Camshaft alignment

    2000 4Runner 2.7L manual
    Taking apart engine due to blown head gasket. Prior to removing the cam shafts I aligned crankshaft to TDC; timing chain mark was vertical, and the "dots" on the Intake and Exhaust camshafts were aligned except they were 1 tooth off.... 2 questions:

    1) When reinstalling should the alignment marks be set correctly or left one off as found? Will one tooth off make a difference in engine operation?

    2) The backlash gear on the Exh. cam is offset so a service bolt can't be installed to keep the load pressure on the spring... is this a big issue?
