Install aftermarket radio in 2001 4 runner

I need help with installing a aftermarket radio in a 2001 4 runner SR5 , on the back of the factory radio is 1 plug and antenna, next to the antenna is a wire ,my question is ,do I hook the radio into that plug or do I remove the factory amp and use the plugs in the amp ,and what's the wire next to the antenna.
