Need more details on the failure:

    Pedal goes to the floor?
Pedal solid but no stopping action?
    Brakes locked up?

Nah, not this time! I'm old school and my mind went to cars designed in the 80's. Plus I had just finished vacuuming my house when I read this, so it was fresh on my brain.

The brake pedal did not go all the way to the floor but certainly went farther than normal without proper stopping action.

^Is the behavior repeatable? Does it happen driving forward?

The behavior did repeat when I placed it in drive. Going up a slight hill to my garage I had to shift to park to stop it. After shutting it down and waiting for a little while I slowly moved it back in the garage and felt the brake engage. I have not attempted to drive on the street since and will probably not until I get it checked out. The master cylinder has adequate fluid.

^Yeah will probably need a tow. Could be gross amount of air in the system or failed master cylinder seals. The latter has not been reported systemically on 5th gen 4Runners, but has been the subject of TSBs and recalls on other Toyota models. When was the last time your brake fluid was replaced?
