PO306 code indicates a misfire in cylinder #6 of your vehicle's engine, the last number indicates the cylinder misfiring.

Have someone check the coils, and or the injectors, they can either test them for cylinder 6 or swap them over to one of other cylinders and see if the misfire follows to that cylinder, replace if necessary.

You didn't say but what engine do you have, a V6?

If so look on your VIN sticker on your driver's door jamb and tell us what the build date is, if it's 12/05 or later the head gaskets are of the newer design, earlier than that date have the old designed head gaskets that have been known to fail and that could be your issue, if the above doesn't solve the issue..

will replace the head gasket, and while I'm at it the water pump and time belt. Keeping my 4runner, only 159k miles, low for a 2005.

The V6 has a timing chain, which is usually good to around 300,000 miles.
The V8 has a timing belt which is good to around 90,000 miles.

But since you doing the head gaskets doing the timing chain is a good idea since it has to come off to remove the heads.

Are you doing the work or getting it done?

I am getting done, have a mechanic for years, he is very reasonable. I live in a subdivision only can do what I can hide in the garage :):)
